I'm planning to extract Polysaccharide from Codium fragile, which is green algae.
I used to use hot water for extraction for 3 hours and then use Ethanol to get them. I tried this way, but It didn't work.
I found out the way that uses Ethanol first for 3 hours and then uses water at room temperature for 18 hours.
Is there difference changing order?
And would you explain why it is plz??

In your question you said that water followed by ethanol extraction did not work, please specify the method you are using to quantify the polysacharides in the extract. Please also say what type of polysacharides you want to extract?

there are several protcols for extraction polysaccharieds from marine algae.
which test you will do after extraction prosses and it is type depends on extraction method due it presented in many forms such as sulphated polysaccharieds
