We have some questions about the storage and stability of cell lysate samples for analysis by the HSP90 alpha Human ELISA Kit (Cat. No.: BMS2090; Thermo Fisher Scientific) and hope that one of you could help us.
We are considering testing cell lysate samples prepared in separate weeks at once. For that, we would like to know your opinion regarding the storage and stability of the samples. The fact that cell lysate samples are prepared in different weeks, could there be losses of HSP90 alpha that could considerably affect our results? Storage at -20°C is indicated in the user guide, however, would it not be more convenient to store the samples at -80°C?

You can store the cell lysate samples at -20℃ but only for 1month or at -80℃ for 6 months without loss of bioactivity. Care must be taken to avoid repeated freeze-thaw cycles. Additionally, while preparing the cell lysates you need to supplement the cell extraction buffer with phosphatase and protease inhibitor cocktail just before use.
I would suggest you store the cell lysate samples at -80 degree C since the samples are prepared in different weeks.

I believe storing samples at -20°C for some time should not be a problem for assays that depend on the recognition of a target protein by antibodies such as the kit you cited.
Storage at -20°C for any length of time is certainly discouraged for other methods, such as assays that measure enzymatic activity or oxidation products (e.g. oxidative stress markers).
