In my dye adsorption experiment the calibration curve is y = 0.046x + 0.018. Now if the absorption value i.e. y = 0.005 for certain concentration it give me the negative value of X. My question is the negative value of X is possible or not and if it is possible then what is the reason for this?
Hello, if your a analysis do not have any relation with regression so I'm agree with below answer.
The y-intercept is 0.018, which is the absorbance in the absence of the dye. If you are getting an absorbance value lower than 0.018, it means there is a problem with your technique or with the equipment, or the noise in the measurement is relatively high.
A few things you can do to get the best results:
1. Make sure your cuvette is very clean. Dyes tend to stick to surfaces.
2. Blank the instrument before each measurement with the solvent.
3. Prepare the standard curve at the same time as you make the measurement.
4. Measure the noise level by taking several readings of a blank.
5. Make sure you are not making your measurement at the same wavelength at which the instrument changes lamps or filters.
6. Use concentrations of dye that give an absorbance of at least 0.1.
