I have fabricated biosensor electrode based on ZnO/Gold/glass. This means that Zno nanorods have been grown on the gold coated glass. I then soldered the copper wire with the electrode on the gold film at one corner.
When I tested by CV (cyclic Voltammatery) it detected the 1mM Ferrocine, but it does not detect the FAD (1mM).
Can you please help me to know where the problem is?
Question seems to be very vague. Assuming CV means Capacitance- Voltage measurement, it is not clear which material is the dielectric. Again, assuming gold coating is one metal plate of the capacitor then what is the next metal plate?
Sir, thank you for the very good explanation. This is the protocol which is followed to check the electrode. Ultimately i want to check this electrode to sense the glucose by using GOD enzyme.
But in case of ZnO nanorods (34nm Dia. and 1um long). Do you think we still need some mediator to transfer the electron to the ZnO nanorod electrode or vice versa to detect FAD.
Did you use the right potential window? The redox formal potential for ferrocene is about +0.3 V vs SCE, but FAD is at around -0.4 V. Try go to negative potentials to see what would happen. Whether or not a redox species is detected by CV depends on the redox species itsefl and the electrode surface property. Any working electrodes have a certain potential window applicable, and thus the electrode material chosen should match the redox potential of targeted species. FAD can readily be detected by CV at carbon materail electrodes such as glassy carbon, graphite and graphene. ZnO-Au electrodes should in principle be fine too.
Sir, Qijin Chi ,thank you very much for the answer.The potential range we are using is 0.2 to -0.8V. Could you please comment about the electrode wire type and diameter. Also please about whether silver paste is good or one can go for soldering.
