1. why we grow citrus rootstocks seed in dark. Is there any advantage to grow it in dark. 2. what possible difficulty I have to face if I grow the grafted plant in solid medium instead of liquid medium. 3. Can I grow it in solid medium by cutting the root.

Citrus grown commercially will reproduce reasonably true to type from seed through a phenomenon known as nucellar embryony. Citrus seeds are grown in dark to enforce emergence of etiolated seedlings only. Because etiolated seedlings means, emergence of true to type plants only.

Thanks a lot for for your valuable suggestion but I would like to know that why the in vitro STG success percentage is higher in etiolated Rootstock in comparison of the rootstock growing in 16/8h light and dark period.
Thats the reason, the harmonal activity is triggered to a greater extent in etiolated seedlings compared to ones grown under normal lights..

many thanks to you sir for sending this paper and i have gone through this is related to regeneration through direct organogenesis. But I want to know that why etiolated tissue give high percentage of success in comparison of green tissue as mentioned various published paper since 1975 to till date.
Thanks a lot Dr. Nemec for your answer if it is so than why people used a drak grow etiolated rootstock for in vitro STG.
