I'm doing experiment on LAB, specifically, lactobacillus plantarum. In my experiment, I inoculated l. plantarum in substrate-containing serum bottles ( in vitro gas production technique) and checked the total gas production, total SCFA concentration and do qPCR to check their initial quantities. The results showed that there was no difference between control and treatments. It means that l. plantarum couldn't survive in the buffered rumen fluid. Is it due to the neutral pH of the rumen? Could anyone explain this for me? Thanks in advance.
Dear Nguyen Qui, if I understood you correctly, you are using just one Lactobacillus species in your research (plantarum), and you are wondering why they are not growing in your substrate!?
Please if it is not a problem for you, could you clarify more clearer about fermentation conditions e.g. substrate composition, growth temperature, pH, time, oxygen content.
And you did qPCR? If you want check the viability of the bacteria better use the simplest colourimetric method.
My suggestions, first you have to analyse the survival ability of your bacteria in artificial rumen fluids and analyse the bacterial growth at different time intervals. Here you can get clear idea and then you can go for gas production analysis.

is filled with bacteria that help the animal digest plants. (In fact, the grass is mainly for the
bacteria to eat. The ruminant animal gets most of its nutrition by digesting the bacteria!) The most

In this experiment, I wanna check if l.plantarum species has probiotic effect on cattle. one of criteria I check is their survial in the rumen fluid. I used in vitro gas production technique. About fermentation conditions, I used elephant grass as the substrate, incubation temperature was 39 celcius degree in buffered rumen fluid (pH=6.8) and in unaerobic condition ( cap & seal the serum right after LAB inoculation). And their survial was checked after 2 days incubation.
After 2 days incubation, I used qPCR to see if there was any quantitative difference of LAB between control and treatment, but there was no difference (P >> 0.05) I wondered why LAB ( l.plantarum in my case) can't survive in rumen fluid?

Thank you very much for your suggestion. By the way, could you please tell me why we need to check their survial in the artifical rumen fluid? What is the benefit? What is the difference between buffered rumen fluid and artificial rumen fluid? Thanks
I think 2 days incubation is too long for L. plantarum. The effect probably detected before 12 hour.
