Both my marker and protein are stacking but they are not going through the stacking gel into the separating gel. Please help. My stacking gel concentration is 4% while the separating gel concentration is 10%.

It may be a problem with the concentration of the gel you used, because the mesh size depends on the concentration of the gel.

Matthew Raj Alias Check the pH of the gels. The difference between the stacking gel and the separation gel is that the pH of the stacking gel is 6.8 while the pH of the separation gel is 8.8.The concentration of the gels is approximately correct.
