Why is SHSY5Y getting killed by protective drug?

I have received passage 29 from our vendor (NCCS, Pune). The cells are slightly different from the previous batch, they form neurospheres, and on adding 12% instead of 10% they show 90% monolayer.

I am working with PN 32, where the cells do not show expected growth reported with an agent as per literature, it is dyeing.

What could be the issue?
