I am staining TLCs in PMA stain which contains ethanol as a solvents. When I dip the TLC plate in it then why is the compound not getting dissolved in that solvent?
I guess the answer is that the dipping is relatively short (2-3 seconds), so your compound, that is adsorbed on the tlc surface doesn't have time to desorb and diffuse significatively. In short it is probably a question of time scale.
Staining solution helps for visibility of your spot, Ex natural product gives visible spot in presence of H2SO4 it means H2SO4 react with your spot and gives visible. if bonding power of silica and spot is greater than bonding power of spot and staining agent than spot will not dissolve in H2SO4 and spot will visible. (staining agent is H2SO4)
