Hi all,

We treated our mice subchronically with PCP and were expecting to see an increased response to amphetamine in a locomotor task. However, we saw the exact opposite. Our PCP-treated mice showed less of an increase in locomotion in response to amphetamine. We have tried 0.3mg/kg, 1mg/kg and 3mg/kg. All three doses of amphetamine elicited less of a response in the PCP-treated animals.

We have tried this twice, using two different treatment paradigms:

1) 7 days of treatment, every 24 hours, 10mg/kg PCP... Testing started on day three after last injection.

2) 10 days of treatment, every 12 hours, 5mg/kg PCP... Testing started one week after last injection.

Mice were 6weeks old at the start of treatment, C57Bl6J, male.

Mice were not on a reverse light-dark cycle and were tested during the day.

Any thoughts would greatly be appreciated!!!
