I synthesed silver nanoparticle from plant. Acording to TEM image some of particular seem dark some of them light. What could be the reason for this?
The light particle maybe a hollow nanoparticle，or its size in the transion direation is evry small (like nanosheet).
Diffraction contrast: Dark particles are oriented close to a zone axis, i.e. there is more diffraction and thus less a reduced intesity of the direct beam.
The reason may be quantum confinement. Quantum effect may have responsible to detect the intensity which is not fair and hence looks light.
Please verify the size effect to support the study using Small Angle X-ray Scattering (SAXS) technique & photo luminescence study.
In your picture there are silver nanoparticles in dark spots in your nanoparticles, and the extract molecules occupy light spots. At the bottom right, the nanoparticle consists only of plant extract (or plant) molecules. To obtain pure silver nanoparticles, you had to rinse the nanoparticles from the extract well. To confirm, determine the content of silver in the nanoparticles and you will see that it is not enough there.
