I am stimulating mice EDL muscle in vitro and the fused tetanus graphs keep having a large bump/ spike in the first part of the graph and then levels off and matches fused tetanus for the remaining part. I have tried adjusting the temperature, increasing the time between tests, increasing the O2 and CO2 concentrations in the solution and, even stimulating at different frequencies and voltage.
