Hi,
I want to detect HIF-1 alpha in HUVECS treated with CoCl2 at 0, 50, and 100uM. I observed two bands at 100kDa (band #1) and 90kDa (band #2) in the treatments and the controls. However, the top band reduced with a corresponding increase in the lower band. I used Anti-HIF 1alpha with a predictable band at 93kDa for WB detection. What modification can cause differences in the two bands? I have attached a tiff file. Has anyone observed this before? And I will appreciate any tips on modifications. Thank you in advance.

I’m also invistigating Hif1 alpha in mPASMC and got the same problem in western blot, but unfortunately, i don’t know why and how to solve it,, i’ll followyour question, might otherone help us
