Good morning, I have a question to ask:
When I add the chitosan (solubilised in 1% acetic acid) to Yeast Nitrogen Base culture medium at pH 3.5, I observed a precipitation of the chitosan.
Could someone kindly explain to me why the chitosan does not remain in solution, even if we are at acidic pH?

Chitosan shows a strong metal-ion chelating ability. The solubility of chitosan is attributed to the presence of amine groups in its structure, which are protonated in acid media, resulting in positive charges distributed along their chains (-NH3+) . Chitosan dissolves in aqueous acid solutions such as acetic, citric, formic, hydrochloric and lactic acids. In the neutralization process, precipitation is observed from pH 6.7, increased up to pH 6.9, and then reduced linearly up to pH 7.4. At pH 5.0 and 6.0, chitosans exhibited good water solubility and zeta potential (ZP) decreased with the MW, whereas the solubility and ZP of the chitosans decreased with increasing MW at pH 7.0
