Third, I would not recommend FRAP assay. This test is performed under much lower pH, ~2, than physiological. Fe(II) is a very active PRO-OXIDANT (https://en.wikipedia.org/wiki/Pro-oxidant). Chelating can reduce this activity.. FRAP measures the amount of Fe(III) reduced by antioxidant. Chelating activity is measured by the amount of Fe(II) bound by a standard chelator. In other words, FRAP measured the antioxidant activity, while other the pro-oxidant activity of Fe(II)

Yurii V Geletii I wont be using it in physiological... Also the obvious answer is that it promotes reduction while metal chelating activity promotes chelation. I wanted to know other chemical reactions involved relating to the effects of iron in both types of assay.

Depending on conditions and antioxidant content numerous reaction can take place. The most evident are Fe(II) oxidation by O2 and Fenton's type reactions. It could be numerous problems with reaction rates. Some reactions might be too slow and you can't find the end points.
