After the addition of 2.5% ninhydrin solution (40% acetic acid/20% ethanol) to glycin samples for the STD curve, the highest concentrations quickly turn purple. However, after boiling for 20 minutes, samples turn yellow instead of purple and develop small dark precipitates. Over time, the color fades, leaving only the precipitates which cause a change in optical density.
