I reviewed above mentioned study where it was observed that granulosa cells show minimal reactivity to PAMPs such as LPS when cultured in serum-free media. However, the paper did not provide a specific reason for this reduced responsiveness. What could be the underlying causes for this phenomenon?
Additionally, when treating granulosa cells with LPS to study inflammatory responses, what is the recommended culture medium composition? Should the medium be serum-free or contain serum, and what are the implications of each choice on the experimental outcomes?

Granulosa cells show minimal reactivity to LPS when cultured in serum-free media because serum contains the required constituents that facilitate LPS-stimulated cytokine secretion. So, granulosa cells need to be cultured in serum containing media to generate inflammation in response to LPS.
When you treat granulosa cells with LPS to study inflammatory responses, you may use serum, follicular fluid, or synthetic supplements in serum-free media depending on the biological question being studied.
Cholesterol supports inflammatory responses to LPS in granulosa cells. For instance, follicular fluid and serum both contain HDL cholesterol but differ in their LDL/VLDL cholesterol content and other respects, including the presence of hormones. Granulosa cells express the receptor for high-density lipoprotein.
