In aqp4-ko and wt mice, we tested a repeated low doses of kainic acid (kA, 10mg/kg, once an hour) and titrated it according to the development of epileptic behavior (racine scale).
In WT mice, the dosage of stage 3 or above was 20 mg/kg.But in the total dosage range of 30 mg/kg, 40 mg/kg, 50 mg/kg and 60 mg/kg, AQP4 KO mice showed only stage 1 or 2 behavioral seizures. Ap4-ko mice exhibited stage 3 behavioral seizures until the total dose of kA reached 70 mg/kg.
Why do AQP4 KO mice need such a large dose to have a Stage 3 seizure?
How does KA metabolize in the body? After intraperitoneal injection of KA, how many KA can penetrate the blood-brain barrier?
