Why B16F10 loss melanin?

I'm working with B16F10cell line... But I have some problem with melanin production. Now (3 passages after thawing) cells are transparent and melanin is only in the medium.. do You know which should be the reason?

Stefania Pezzana sorry this response is coming late. However, melanin production by B16 cell lines depends on the number of previous passages it has had.

If you recently bought the cells and use complete DMEM to grow them, you should not have trouble with pigmentation.... Alot of suggestions has been given regarding your current situation. You can read here:
