the closer the sample is to a ladder, the more curved line i get. I run the samples in 0.7% agarose gel for 30 minutes, 150mV. Why does it happen and how can i improve my gel?
It is most likely one of two things. It could be the gel was not properly covered with buffer such that the current did not flow evenly across the entire gel (or one buffer compartment wasn't quite full). Or (and I think this is more likely) your samples have a lot of salt in them. You can do an ethanol precipitation to clean them again or maybe try to just load less. You have so much material in your samples try loading about 1/3 the amount and see if the gel is more clear then.
That voltage is rather high, you're risking melting your agarose gel. I'd recommend turning it down to 120 or lower.
I'd also recommend that you wash the gel rig, combs, tray, etc. and rise really well with distilled water. Dirty gel combs with a build-up of salts can also cause this sort of bending/smearing.
