I observed blue with shades of red colours with irregular shapes while viewing a pure bacterial culture under a light microscope. What could be the reason?
In liquid preparations, probably air bubbles. In air-dried smears, residues from the medium. And of course, if the microscope is not properly focused, you see rings that are either blue or red. Proper focus is achieved just between the red and blue.
Sounds like diffraction of the light into red and blue colors. Could be from smears on the microscope lenses (very possible) or from material on your slides. I'd suggest carefully cleaning the lenses, objectives, condenser, etc. on your microscope and see if that helps. Could also try viewing your slide with another scope and seeing if it looks any different. Good luck!
