I am working on transgenic rice. We have standarised the regeneration and transformation protocol for it. But I could see two Albino plants that are regenerated from calli. Can you please help me to short out the issue. For your kind information I want to mention that other plants regenerted with same media and hormone componets including same light condition have not shown such abnormalities. The origin of this behavior might be multiple. Considering that you don't direct the insertion to a specific place in genome, it can leads to the appearing of this phenotype. The tissue culture by itself could be the cause of variations regarding the original patterns. This phenomenon was described by Larking and Scowcroft in 1981 and was named somaclonal variation (see Larkin, Philip J., and Wv R. Scowcroft. "Somaclonal variation—a novel source of variability from cell cultures for plant improvement." Theoretical and applied genetics 60.4 (1981): 197-214).

I dif not searched about ricr, but as i knew in some ornamental plants same African violet and Coleus, when you propagated them invitro, their color changed, as i remember in coleus even i saw albino...if you search these subjects, you find, but i must check my articles from my tablet, later when i check, i send that albino coleus made from tissue culture..
I just probably say this reason, because i dont have information about rice.

Jyoti Prasad Kakati Dear I think this due to plant growth regulator influence. I think if you are not using Myo-Inositol in regeneration media than try this in 100 mg/l concentration. And if already using, try to enhance the amount. MI plays a critical role in in vitro greening of leaves.

An albino plant, generated from a transformation event in tissue culture can easily arise by loss of plastids in an early stage of regeneration, as transformation occurs at the single cell level. Plastids/chloroplasts are required by a cell to rejuvenate and produce photosynthesis products, eg sugars and other derivatives. If you have done propagation with sugar in media then there is no selection for plants to photosynthesize and regenerate, thus the plastid-less plantlets may be able to survive.
The transformed rice seeds having kanamycin resistant gene as selection marker can produce albino plant in vitro. This is not unusual as kanamycin at particular concentration blocks ribosomal RNA synthesis in plastids resulting in no synthesis of chlorophyll.

I also observed the same phenomena in Bamboo also, its nothing but a Somaclonal variation. Generally these plant are not survival in nature, this is my observation in the filed.

Its a bit difficult to conclude on the main cause of Albinisms in Rice.
Yes, it may be due to loss of plastids or somaclonal variation. In our research team we get a lot of Albino plants in the rice anther culture. We are now finding out if there is any genetic mutation associated with it.
What variety of rice did you use? Was it PB1? I have seen similar things in PB1 rice. I am not sure what the reason is but I remember this phenotype was specific to PB1 rice. I was growing 8 other varieties and none showed this phenotype. Moreover, the rice was being grown without any selection.
