Why exactly are you trying to reduce Ace-CoA formation? I guess is for avoiding excesive production of acetate. Is that the case?
Hi Karla, Thanks for your inquiry. My concern with Acetyl-CoA is that i want to redirect the carbon flux to lactic acid but have low acetyl-CoA
The flux towards Ace-CoA is higly regulated and in a WT E. coli; so this flux is really favorated in all the conditions. If you are interesed in perform some mutations in the strain to produce lactate I suggest you read this paper http://www.ncbi.nlm.nih.gov/pubmed/19471981, although this paper is a mineral media, I think it could work fine in your rich media. On the other hand you forget to mention if your culture es aerobic or anaerobic. I guess is anareobic right?

Traditionally, acetate excretion in E. coli has been attributed to an overflow metabolism phenomenon. Hence, fed batch culture strategies are recommended. Thank you Daniel and the others for your suggestions. Happy to tell you that i got a mutant strain that produces extremely high levels of lactic acid and now considering to regulate its production
