Which solvent system is recommended to extract betanin from crude extract using column chromatography? Any other suggestions on extraction methods that can be easily set-up from scratch are also welcome.
you can try a mix of methanol: water (8:2) with silica gel column. Try acidifying the column a bit using either formic acid or Glacial acetic acid. You can also try reverse phase chromatography.
Have a look at the link given for a paper I found, I hope that will be of your help
You can try and see what will be the effect of ethanol and methanol on your compound mixture which you are seeking to separate. The only difference between these alcohols is on their eluent strength. Methanol, in general, has a higher eluent strength (0.95) than ethanol (0.88). That's why methanol is preferred.
