I would like to use the immunogold labeling method and TEM and SEM for localizing. All the previous works are done on localization of enzymes. I cannot find any previous works done. I need your tips, thanks in advance.
If you look Table 1 of the review below, the writters identified somme publications by an I for Immunogold Labeling. I hope this will help you.
Roze, Ludmila V., Anindya Chanda, and John E. Linz. “Compartmentalization and Molecular Traffic in Secondary Metabolism: A New Understanding of Established Cellular Processes.” Fungal Genetics and Biology 48, no. 1 (January 2011): 35–48. doi:10.1016/j.fgb.2010.05.006.
Since you are looking for a compound and not a protein/ enzyme, I highly recommend that you use high pressure freezing and freeze substitution to prevent loss of aflatoxin b1. HPF will be able to halt the metabolic process faster than normal chemical fixation and freeze substitution will prevent loss of aflatoxin b1. I also suggest you a lowicryl based resin for resin embedding and uv polymerisation at temperatures below 0 degrees. You can then section and do immunolabelling. Ensure to include the correct controls.
I have had similar experience in localising caffeine within tea leaves. I suggest you read it as I am sure it will help. It is on my research gate page.
