Besides the literature I think u have to see that how much debris is in the sample then on the basis of that u have to descide

As if sample contains large amount of debris u have to use nitric acid digestion method

And if less u can go for H2O2 and Hcl method

Bcoz if sample r not properly digested it would hinder the microscopic image

Ajay Singh Rana Sir can you share the protocol of HCl and H2O2 digestion method as there are so many available in the literature so i am little bit confused.

Please try all the method like

Hcl and H2o2

Hno3

Kmno4

Lugol iodine

The results should check on the basis of clarity of diatoms, transparent digested material, diatoms should intact without much destruction or eroding, the method should not be lengthy as also cause loss of diatoms during processing

So then set your protocol
