Which is better EDTA or heparinized tubes for mouse blood, I tried EDTA but it causes clotting sometimes although I put 1-2 ml blood only in the tube. And I tried heparin but it caused hemolysis . I need healthy WBC and I will also perform some ELISA parameters and complete blood count. What is the best speed of centrifuge (rpm) that I should use to separate plasma from cells without damage? what is the duration?
Heparin is normally recommended, but I've used EDTA with no problems in some ELISAs. To prepare plasma you can centrifuge 15min, 1000g at room temperature to obtain platelet rich plasma, and if you move the plasma to a new tube and centrifuge 10min, 10000g at room temperature you get platelet poor plasma that you can store at -80ºC and use for ELISA.

EDTA is better tube for blood cell count because EDTA about 4 hours morphology WBC, RDC and platelet is good . A cause cloting is possible your not mixing for 8- 10 times correctly.

Check that the ratio of anti-coagulant to blood volume is within recommended limits. What type of tubes are you using to collect the blood? If you are needing to do functional assays on the WBCs, then heparin is best.

Heparin per se is an important stimulator of blood ROS Generation. For this purpose citrated blood is much better, EDTA acts like citrate+calprotectin.
