As IL-4 primes macrophages toward an M2 (alternatively activated) phenotype, and you should see IL-10, TGFbeta, but NO IL-12. In general, whatever cytokines you can imagine being produced by an immunosuppressive macrophage, they produce it. There is a lot of literature out there, describing differential cytokine,MMPs or CCL. Main question is whether you want to prime THP-1 for example (or any other macrophage line), or use primary cells. In the second case, there are also loads of changes in CD marker expression involved.
