As you pointed, a wide range of % has been reported in the literature. depending on the type of chitosan you are using (Mw, solution pH, etc) it is not easy to decide unless to optimize the process. In our experiments we selected a range of 1 to 10% (w/v) TPP solution, which chitosan solution added drop wise to it, followed by stirring and centrifuge. chitosan was also in the range of 0.5-2% (w/v).

You can use the following instruction to obtain chitosan nanoparticles with ca. 100 nm:

1 g chitosan was dissolved in 100 mL acetic acid solution (1% w/v) overnight. Then, the TPP solution (1% w/v) was added dropwise to filtered chitosan solution under vigorous stirring a for 40 min. The prepared suspension was subsequently centrifuged at 10,000 rpm for 20 min at 4°C.
