I have a question concerning the elution step during the isolation of cfDNA with QIAamp Circulating Nucleic Acid Kit. We care a lot about the high efficiency of isolation of cfDNA and for this reason, we decided to eluate twice. Last time because of the high concentration of cfDNA in the first (2,6 ng/ul) and second elution (0,692 ng/ul), we made the third elution to isolate as much DNA as possible. The concentration was the same as for the first elution(2,6 ng/ul). Is it possible? We are using Qubit Fluorometer 1x dsDNA high sensitivity assay to measure DNA concentration, which is specific for DNA.
