This bacterial strain comes from Life Technologies and appears to have been designed with a resistance to ccdB (ie it is a strain for propagating Gateway entry and destination vectors). Are you actually working with the Gateway recombination technology or are you just looking for a strain that will give you high transformation efficiency?
Thank you for your response. I am working with Gateway recombination technology. I need to make a lentiviral destination vector, that is why I am looking for DB3.1 which is recombination deficient and ccdB resistant. Does Promega have something like this. Thanks
Thank you for your reply Sathyan. no, we do not have a ccdB resistant bacterial strain for propagating Gateway vectors. It appears that Life Technologies have a One Shot ccdB Survival Strain. See http://www.lifetechnologies.com/order/catalog/product/A10460. That is what I would recommend you use.
Life Technologies indeed provides the One Shot ccdB survival strain as an alternative, however we've noticed this strain doesn't work equally well for all plasmids. With the DB3.1 strain, so far we never had any issues.
