I sent my revised paper by 25 April to a Springer journal, till today there is no progress on my paper the status of the paper is still " Reviewers assigned" . Do you think that i can contact Editor in chief after 4 months of waiting?, since the handling editor doesn't reply to my queries.

Yeah, you can contact the Editor-in-chief and explain your concerns. Editors have interest in providing good and reliable services to their authors and so I believe you will get a response.

I think yes. you must write an inquiry to editor in chief about your manuscript's current status. you can use this sample letter:
Dear Mr./Ms. XXX [Editor's Name],
I have submitted my revised manuscript titled XXXX [manuscript id: xxxx] to your journal via the online submission system on dd/mm/yyyy [date of submission].’ However, the status has remained unchanged ("Reviewers assigned") ever since.
I would be grateful if you could let me know whether there has been any further progress on my submission.
