I am thinking of measuring the degree of substitution of esterified polysaccharides by alkaline hydrolysis, followed by extraction with water-immiscible solvents such as ethyl acetate. This is an approach I have used successfully before. Unfortunately our lab does not have a speed-vac, which would enable me to dry down the sovent extracts before RP-HPLC. Would SPE cartridges be suitable, and if so, which type?

The carboxylic acid substituents are mostly non-polar (i.e. oleic acid, lauric acid, ferulic acid), but there may be some with cationic or anionic substituents (i.e. betaine, succinic acid).

The elution solvent for the HPLC system would probably be acetonitrile : water.
