I’m working on mouse cardiomyocytes and I wonder if different energy providing substrates would perform differently in cardiomyocytes in matter of force development, excitability, viability. I tried to image ATP by indirect measuring with Magnesium-Green, but could only find effects due to Na-Azide, but not due to feeding different substrates. Next I tried to take spontaneous calcium signaling as measure but the gained results were rather inconsistent. Now I wonder if different energy substrates would have on effect on force development and/or heart performance. Please let me know if you have made some experience or if have a better idea.
