I want to make sections of mangrove roots to search for mycorrhizal symbionts. Which solutions are usually used for the fixation and dyeing of this kind of samples ? The fixative agents may not interfere with the dyes and I would like to distinguish the wood from the hyphae clearly.
@ Anton, to observe structure within the root, it is necessary to clear cortical cells of cytoplasm and phenolic compounds, and then to differentially stain the fungal tissue. Phillips and Hayman (1970) developed the method to visualize AM fungi in roots using trypan blue in lactophenol, but the use of phenol is later discouraged (Koske and Gemma, 1989). The KOH is used in clearing agent for non-pigmented roots, but H2O2 (Phillips and Hayman, 1970) or NaOCL (Graham et al., 1992) may be used for pigmented roots.
Also, make sure you reserve uncontaminated new reagents for your use only so that you can be confident of your results
