The age of the rat to be used depends on the method you want to use to induce type-2 diabetes.
About 3 month-old rats can be used in the case of Streptozotocin-nicotenamide-induced diabetes; about 2 to 4 month-old rats are normally used in High-fat-diet and streptozotocin-induced model, as well as High-fat-diet and low-dose streptozotocin – induced model.
Type 2 DM can be induced in rats by I. P. injection of streptozotocin to neonatal rats. Normally, a single dose of streptozotocin is administered in the first day of life (100 mg/kg) or on day 2, 3 or 5 (120 mg/kg). After 8 to 10 weeks of age these rats manifest basal hyperglycemia, impaired glucose tolerance and loss of beta-cell sensitivity to glucose.
For the above models both Wister and SD rats can be used. SD rats may be easy to handle. Wister rats have been used more often in the published papers.
There are a few spontaneous or genetically derived obese type 2 diabetic rats. These include Zucker diabetic fatty rats (about 4 weeks of age) and Ostsuka Long-evans Tokushima fat rat.
