We want to study the antimicrobial potential of medicinal plant extracts. Kindly suggest the precautions for antibacterial and antifungal activities.
The main concern is that an extract may be not just antimicrobial but antiseptic, killing everything. Your first hint will be when it kills both bacteria and fungi. You should plan to counterscreen for toxicity to a mammalian cell line in culture.

Thanks for guidance but i don't know about the methods for counter screen to a mammalian cell.
One method is to treat a culture of cells, such as an immortalized human cell line (e.g. A562), with a set of serial dilutions of the extract in a 96-well microplate for a day, then measure the metabolic activity of the cells using a tetrazolium dye assay.
Use only standard strains for your animicrobila or anti-fugal assay.

1. First collect the standard strains from a culture collection centre.
2. Prepare the extracts both aqueos and solvent based.
3. I advice to follow a disc diffusion method first through which you can analyse a large no. of extracts vs. microorganisms.
3. Follow a microplate dilution method through tetrazolium assay.
4. Indirectly you can find the way of action of the extract comparying activity with standard antimicrobial discs.
5. Run a TLC of the extracts.
6. You can run a Oxanography method to know the compound active in the extact mixture.
7. You can think for isolation nad characterization of the compound
