More the concentrated is your sample less it requires for the analysis. Although all it depends upon concentration of the sample even then minimum 75% vial of capacity 2ml should be filled for the best results.
In addition to the vial fill, concentration, piercing etc. precautions which are very contributing in analysis, check the limit of detection of your machine and be well above it, x10, if possible. Go through the manual and see the recommended operating procedures along with other indications on fruitful analysis. The manual can also be had online.
