Actually am working on marine bacteria, extraction of their crude extract and analysis for antimicrobial activity. One of the isolate/bacteria shows significant antibacterial activity upon primary screening (point inoculation) as well as on well diffusion method. But upon extraction of crude extract in different organic solvents of different polarity index, none of them show any activity. Even i have treated its supernatant with proteinase K for nature determination and the metabolite is not proteinaceous in nature. Suggestions are highly appreciated. Thanks
The antibacterial substance may be unstable under the extraction conditions, or it may be a mixture of substances that are being separated by the extraction procedure.
May be different organic solvents that you used it are not suitable for extraction.
Or may be as Dr. Adam tell you it may be a mixture of substances that are being separated by the extraction procedure.
