I am interested in designing an experiment involving the quantification of M. cerebralis actinospores (TAMs) released from infected T. tubifex under laboratory conditions. I have seen other papers describing using microscopy to detect and count TAMs in water samples, although I want to be sure that I have the correct methods. Will standard light microscopy work to detect TAMs? What magnification is best? If standard light microscopy will not work, what other techniques are required?
