I am trying to study controlled delivery of cisplatin. But the problem was that I am unable to draw a calibration curve. Initially I took cisplatin in little amount of DMSO and then add water to volume make up. When I tried to take uv, I found no absorbance peak. Then how do I determine LDE, LDC as well as study the release profiles?
Hello! I think that simple UV detection is not a best method for cisplatin determination. Take a look at this paper http://www.hindawi.com/journals/isrn/2013/936254/
If you want to develop a simple and sensitive method for drugs with low sensitivity in UV region then it is always better to form derivatives those may be colorful or colorless, but should have strong absorbance in the UV/Visible region. Then validate the method as per ICH Q2 (R1) guidelines.

when I am taking Cisplatin in DMSO-water mixture, it is not showing any absorbance peak in UV-vis spectroscopy..even in only water medium also (that is may be due to insolubility in water)

Finally I have done this. Thanks given to Dorota Danielak. Shortly, I have taken DMF solution of cisplatin and to it DMF solution of o-phenylene diamine was added and heated at 90 degree celcius. UV data of the resulting complex was collected. I got satisfactory results in this method.
