In my cytogenetics laboratory, we usually get good mitotic index and good chromosome spreads. Recently there is some small thread like particles that is visible after the addition of first fixative. I have attached the images.
What could be the reason for such particles ? Need help to understand the source.

It could be some problem with the giemsa, or some debris in the slide. It is totally irrelevant for the diagnosis. We have this Kind of debris in our laboratory, with out another problem.
This is not a real problem to concern.

Is this visible in unstained preparation as you have mentioned 'after first fixative'? Or is it after the GTG banding? Is this a cancer tissue - if yes, these look little bit like 'double minutes' but not well defined. Do you see same chromosomes involved in this structural aberration in all mets or is it random? Need more details before considering it as artifact.
Associted artifacts often found in metaphase spreads, these artifacts can be cleared by treating slide with 1N HCl for about 40 minutes.If these residues are bits of chromtin that may be due to over treatment of material with acetic acid while preparing slides becuase over treatment leads to over spreading of chromatin .If these artifact appear only after staining it may be due to residues in giemsa.or if these fragmented structures maifest in G banded stages ,it may be due to over treatment of chromosomes with trypsin .

Thank you all...this is peripheral blood karyotyping. We do treat the slides with HCl and casting the slides the next day of harvesting has shown some improvement.
After air drying the slides for a day before straining, wash in water by dipping to remove acid
