I have gone through some of the methods used for finding the mode of action on pathogens such as cell viability assays, cell membrane permeability etc. But I would be grateful to anyone, that if you could just explain me the purpose of experimenting the effect on respiratory chain dehydrogenase enzyme activity of pathogenic bacteria as a test for mechanism of action for a particular drug/ plant extract?

Functional respiratory chain is required for full virulence.
Probably the entire respiratory chain consists two enzymes, a type 2 NADH dehydrogenase (NDH-2) and a cytochrome bd oxygen reductase.
So respiratory dehydrogenases feed electrons into the respiratory chain, and respiratory oxygen reductase reduce oxygen to water.
