I am using C18 250x4.6 mm. After injecting the column with Poultry feed vitamin premix samples, I noticed a high elevation in the column back pressure. My mobile phase is composed of 99% Methanol and 1% Water. Does anyone have any suggestions?
Hi, The elevation in column back pressure may be due to deposition of analyte or buffer etc. You can wash the entire HPLC system with your mobile phase solvents. wash the column 1st with methanol and then with water for a long time. you can also try hot water for washing. regards.
If you are going to take Avtar's advice be sure to take the column offline. C18 columns do not LIKE 100% Water (it causes column collapse and will kill it),
