what step should i take from step one to produce significant results,especially the how to mix culture media and PEG?
There are two most important factors for concerning to mimic the natural drought tolerance, they are factors of water and pressure. And you mentioned the water factor, and PEG is a good choice. So the concentration of PEG for screening is the most important for you. For me, I will read some related papers in sesame, and get a proper range of the concentration of PEG, then do a pre-experiment to see the exact concentration of PEG for your experiment, and the concentration of PEG need to be used for discrimination of your landrances (simply say that you can see the different phynotypes of your landrances under that concentration of PEG). And you can also choose the classic concentrations of PEG for screening for drought tolerance in sesame.

Go to Plantstress.com. Click on 'Methods' where you will see my comment on working with PEG. PEG in nutrient solution giving 3 or 5 MPa of potential might be appropriate. Before you lay out the experiment try the method with one variety and several PEG concentrations to get the hang of it and find the best PEG concentration for sesame.
PEG medium is best choice for screening at germination and measurement of coleoptile length at seedling stages might provide you information about drought tolerance in sesame germplasm.

I agree with Avta PEG based drought screening approach is ideal for germinating seed for so they will be no need to add culture media but for plants sorbitol will do then you can use Hoagland nutrient solution as a culture solution
I agree with tashings that PEG dissolved in any nutrient solution is very convenient to carry out drought stress experiment. To chose the PEG concentrations, read some publications on that and have a trail experiment to decide with sesame.
