Again this is very simple. Just extract the chlorophyll in acetone or ethanol and estimate it (using standard chlorophyll) either using HPLC or GC with ECD. This will not quantitate the different types of chlorophyll. For that purpose, the simple way is to run a preparative TLC and then collect the different fractions separately and then analyze it on HPLC.
