What is the optimum concentration (and process)of etomoxir to block CPT1 activity on A549 cells?

I did both CPT1 and FAO assay with etomoxir conc. of 50 to 200uM in A549 cells but it didnot show any inhibition of CPT1 or FAO, Why?

Dear Dr. Niroj Kumar Meher

You may have to increase the concentration of etomoxir. Increase the concentration of etomoxir to 500µM. Use the range of 500 to 1000µM instead of 50 to 200µM. The range of 50 to 200µM for A549 cells may be too low a concentration.

I have attached a paper below wherein the investigators used 500µM which could inhibit FAO in A549 cells. See Fig 5A and 5B.
