I need to test the toxic effect of 2 different compounds on cell-lines (in 96-well plates)
For each compound, I have 6 different concentrations.
And in two different conditions: in 1° an amino-acid and glucose-rich culture medium, and 2°/ a poor culture medium.
The objective is to check:
1° the effect of the 2 toxic compounds (and if one is more toxic than another)
2° is there a threshold where the toxicity begins?
3° can the rich culture medium confer a relative protection compared with the poor medium?
And the endpoint was the cell viability measured by MTT-assay.
I would like to know the most appropriate design (statistic test) to analyze this.

If you need to compare the toxicity of two compounds with each other and with the control in the MTT test, it is better to use the ANOVA assay. First, you need to compare the actions of the compounds in the medium with serum, and then compare the actions of the compounds in the medium without serum.

Marius Baguma, assuming that the data is normally distributed then yes it seems that you will need 2-way ANOVA. Good luck.
