I try to solubilize a powder with a mixed solution of tween20 and ethanol for only two injection (IP) in 7 weeks old BALB/c mice. The injection volume is 100µL. Can you help me ? Regards and Thanks
some drugs that are not soluble in tween 20 and ethanol are soluble in bicarbonate du to a change in the pH. Have you tried this option? if your drug is not solubilized and you carry out an ip injection, you risk that some of your compound does not reach the peripheral blood in a correct concentratrion.
I apologize for the delay in my answer. We knew that our compound can be solubilized in ethanol and Tween20. We made different tests by making different ratios between ethanol, tween and water and we had good result;
Personally I don't know your proposition, we have already tested it and I keep this solution for other drugs. Concerning the potential mistake of an incorrect concentration injected, this is why I posted this topic.
This question has already been solved. It was treated on the following topic on researchgate:
What's the maximum volume of ethanol a mouse can take? (7 years ago)
from Loredana Vesci :Ehanol is used at a max concentration of 5% for formulations with cremophor or DMSO, for instance and it may be used every day. This suggests that a mouse of 20 gr can receive a maximum amount of ethanol (10 mg) if you use a standard volume of 10 mL/kg.
