What is the final concentration of ADP required to stimulate state 3 respiration in Clark electrode?

We are using Oxytherm instrument from HansaTech. If anyone is using the same instrument, please let me know the final concentration of ADP required to stimulate STATE-3 respiration in Clark electrode. We are adding 25 microlitter of 30mM of ADP stock solution to make final concentration of ADP of 0.5mM in the chamber of 1.5ml to stimulate the Clark electrode. However, we are not getting any change in rate after adding this concentration. We are using 7 mg/ml of mitochondrial concentration for this experiment.
